Shimadzu’s New Nexera X2 Achieves World-Leading Sensitivity and Superior Separation Across Entire HPLC/UHPLC Range

Shimadzu optimized the Nexera X2 to enable analysis at pressures up to 19,000 psi. Without compromising performance and data quality, the maximized pressure over a wide flow range enables various types of analyses. Nexera X2 autosamplers provide near-zero carryover, support ultra-fast analysis with the world’s fastest cycle time (14 secs), and deliver excellent injection reproducibility down to 0.1 ul injection volumes.

Users can configure the Nexera X2 system with a variety of solvent delivery units and column ovens to meet any laboratory workflow. In addition, its new mobile phase blending function automates mobile phase preparation tasks for greater efficiency.

New core features of the Nexera X2 include:

SPD-M30A Photodiode Array Detector: The new SPD-M30A offers superior sensitivity and resolution, achieving a 0.4x10-5 noise level through the adoption of the total reflection capillary flow cell (SR-Cell), new high-brightness lamp and optimized signal processing circuit. The SR-Cell maintains superior detector sensitivity while minimizing peak dispersion in the cell. This maximizes peak separation through excellent peak shape. A high-sensitivity version of the cell is also available. Featuring an 85-mm optical path length, this model quintuples the sensitivity of the standard SR-Cell, enabling detection of ultra-trace impurities.

Intelligent Dynamic Range Extension Calculator (iDReC, patent pending): The newly developed iDReC function in the SPD-M30A extends the dynamic range so low concentration (trace or impurity compounds) and very high concentration samples can be accurately analyzed in a single injection. Accurate calculation of flat-top peaks is possible with a linear calibration curve, making the traditional limitations of a detector’s linear range obsolete. Users can combine this with the high-sensitivity SR-Cell to extend the measurement concentration range by a factor of 50.

Intelligent Peak Deconvolution Analysis (i-PDeA): The newly developed i-PDeA extracts a single peak from co-eluted peaks and quantitates it by exploiting the differences in spectra between each compound. The i-PDeA enables users to visualize and detect a minor single impurity even when the impurity is co-eluted with an analyte. It also facilitates the separation of hard-to-separate peaks on the column through computer processing and deconvolution of the spectrum information, which reduces the effort required to investigate separation parameters.

Temperature Controlled Optics (TC-Optics): The SPD-M30A also provides unrivaled baseline stability and near-zero baseline noise due to TC-Optics. The SR-Cell has an optimized heat exchange inlet pipe with low external dispersion. With temperature control, the SPD-M30A achieves superior separation while enabling faster stabilization after startup. These advances shorten wait-time before each analysis and provide more reliable testing by keeping a stable baseline against shifts in room temperature. Further advances in the optics platform allow for a vast increase in spectral resolution.

Automated Solvent Blending: The modular nature of Nexera X2 allows a solvent blending function that can mix solvents according to desired ratios. For example, a high-pressure gradient can be modified with automatic addition of modifiers for LCMS ionization, ion pairing, or pH gradient formation for optimizing mobile phase conditions in method development.

The Nexera X2 system’s modular design allows desired system configurations based on application needs. Equipped with the new SPD-M30A and i-PDeA function, the Nexera SR configuration combines high sensitivity and high separation. It can also include a Nexera Quaternary pump, a four-solvent gradient analysis system, which is compatible for both HPLC and UHPLC analysis, allowing for easy method transfer.

Another configuration in the Nexera X2 Series is the Nexera MP. A front-end HPLC for all LCMS platforms, Nexera MP utilizes the multiplate SIL-30ACMP autosampler, which features an ultra-fast injection (7 sec), near-zero carryover and continuous analysis of up to 2,304 samples, making it ideal for high-throughput multi-sample applications. The series also offers the Nexera Method Scouting configuration, an exhaustive method development system that allows comprehensive method scouting using up to 96 combinations of mobile phases and columns.
