Features of GlycoWorks RapiFluor-MS N-Glycan Kit
Streamlined Workflow            GlycoWorks RapiFluor-MS N-Glycan Kit reduces complicated, time consuming sample preparation.

  • Easy-to-follow protocol
  • Detailed tips and tricks provided for adaptation
  • Easily train non-glycans experts
Fast Sample Preparation            The entire process, from glycoprotein to ready-to-analyze sample, can now be completed in less than 30 minutes*. This is made possible by fast deglycosylation combined with instant labeling.

  • Increase lab efficiency
  • Quickly analyze screen samples and provide feedback in a timely manner
  • View how it is done with the online demonstration video
Unprecedented Sensitivity            RapiFluor-MS is a reagent comprised of an NHS carbamate rapid tagging group, an efficient quinoline fluorophore, and a highly basic tertiary amine for enhancing MS ionization. As an NHS carbamate, unlike alternative rapid tagging reagents, it hydrolyzes over time to generate carbon dioxide and a corresponding amine. This makes RapiFluor-MS labeling a self-quenching reaction.                         Enhanced chemical properties for glycan analysis:

  • Rapid tagging
  • Efficient fluorescence
  • Enhanced ionization efficiency
One Kit, 96 Samples            The GlycoWorks RapiFluor-MS N-Glycan Kit provides enough materials to process 96 samples, 24 at a time.

  • Deglycosylation Module: Rapid PNGase F, customized buffer, RapiGest SF Surfactant, and the Intact mAb Mass Check Standard as a control to benchmark the protocol.
  • Labeling Module: RapiFluor-MS Reagent and Reagent Solvent.
  • Clean-up Module: GlycoWorks HILIC µElution Plate* and the GlycoWorks Elution Buffer and Sample Diluent.
  • Sample Collection Module: Tubes, caps, collection tray, and waste tray needed to handle samples.
Characterization Made Simpler            RapiFluor-MS provides increased fluorescence quantification along with unprecedented mass spectral signal intensity. Low abundance glycans can thus by easily assigned and characterized by serial fluorescence and mass spectrometric detection. Less sample can be used while still obtaining increased sensitivity.                        When labeled glycans were analyzed by HILIC chromatography, RapiFluor-MS produced the following results compared to the following:

  • Commercial rapid tagging analog of aminobenzamide (AB); 2x greater fluorescence signal and 1000x higher MS signal
  • Conventional reductive amination with 2-AB; 4x greater fluorescence signal and 160x higher MS signal
Assign a Mass to Every Glycan Peak            The ACQUITY QDa Mass Detector routinely detects RapiFluor-MS labeled N-glycans.

  • Equivalent FLR and MS response all within a single run, with the added benefit of mass confirmation.
  • Enables higher throughput targeted glycan profiling for better informed process development.
  • Easily integrated with existing GMP compliant-ready UPLC workflows with Empower Software.
