iPSC Reprogramming of PBMCs using the L7™ PBMC Reprogramming Bundle

Lonza’s Pluripotent Stem Cell Innovation Team has developed an optimized protocol for iPSC reprogramming starting from peripheral blood mononuclear cells (PBMCs) using the 4D-Nucleofector™ System in combination with episomal vectors.     
     
iPSC reprogramming of PBMCs and in vitro differentiation into embryoid bodies. After 14 days of differentiation in the presence of FBS, differentiated cells were tested for the expression of proteins of representative cell types of the three germ layers. Top right: Ectoderm, stained with Tuj1 antibody against neuron-specific beta-tubulin III. Bottom left: Endoderm stained with AFP antibody against alpha-fetoprotein. Bottom right: Mesoderm, stained with SMA antibody targeting smooth muscle actin.

Components required for the iPSC reprogramming protocol:     

  • Human PBMCs: PBMCs isolatedfrom patient material. In addition, Lonza's cryopreserved human PBMCs are suited as positive control donor for reprogramming.           * NEW! L7™ PBMC Priming-Recovery Kit: The kit contains a basal medium that needs to be supplemented with recommended growth factors and cytokines (not provided). The supplemented media supports priming of PBMCs towards erythroblasts prior to reprogramming as well as a four-day recovery phase post transfection. For optimal programming results, the kits also includes L7™ PBMC Reprogramming Enhancers A and B.           * P3 Primary Cell 4D-Nucleofector™ Kit: For efficient transfection of episomal vectors into the primed PBMCs we have established the 4D-Nucleofector™ X Unit with P3 Primary Cell Solution.           * L7™ hPSC Culture System: Seamless combination with our for feeder- and xeno-free iPSC culture system
