Dextran Calibration Ladder
In order to take full advantage of the BEH glycan column, Waters created a line of labeled Dextran Calibration Ladder Standards to assist glycan profiling. Glycan profile obtained from UPLC-HILIC/FLR systems can be calibrated against the labeled dextran ladder and assigned with glucose unit (GU) values. Waters Labeled Dextran Calibration Ladders are different than the existing commercial offerings. The average molecular weight of the glucose homopolymer is higher (~4,500 Dalton), therefore, the workable GU value range is twice as much as the other dextran ladder standards; the observed GU goes from 2 to 30. This feature improves the large glycan retention time assignment.

Waters Ammonium Formate Solution
Quality of reagents is critical to mass spectrometry analysis. P/n 186007081 is a concentrated version of the 50mM Ammonium Formate buffer that is recommended to be used with the Biopharm/Glycan system, using the UPLC H-class and the ACQUITY UPLC BEH Glycan Column.
This buffer contains 10 ml of 5050 mM ammonium formate in a water w/ 3.8% formic acid matrix
This allows the end user to directly pour the concentrate into a 1 L bottle of LCMS grade water for the correct concentration of 50 mM at a pH of 4.4
Raw materials are all QC’d to ensure high quality MS friendly reagents which minimize adducts
The HILIC Advantage:
Hydrophilic Interaction Chromatography (HILIC) can be used to successfully improve retention of very polar species. This technique is rapidly growing for this application area. One reason for this is that this mode of separation is well suited for glycans due to their highly polar nature. The GlycoWorks HILIC SPE provides an efficient and reproducible mechanism for effective glycan cleanup and provides quantitative and consistent recoveries for a diverse range of N-glycans. Along with the GlycoWorks SPE Reagent Kit with a customized elution buffer this provides a very reproducible cleanup capturing species up to tetrasialyated.
