No. After filling the 25ul loop rest of things goes to waste

However, when I load a 200 ul sample volume my peak area is 2x the size of that of 100ul.

If the rest goes to waste I should not be seeing my peak area double?

The Autosampler Model is Dionex AS, loop sizes are 25ul

Definitely there should not be such kind of problem. I think you should check the loop once again or check the injector linearity.   Also, did you get reproducibitly for the standard?

that means autosampler not taking the exact injection volume

I think you have to program into the auto sampler the size of your sample loop if you have this kind of problem.
