Senescence associated ß-galactosidase (SA-ß-gal) is a widely used biochemical marker for assessing cellular senescence. SA-ß-gal can be detected by 5-bromo-4-chloro-3-indolyl-ß-D-galactopyranoside (X-gal) at pH 6.0, which inhibit lysosomal ß-galactosidase activity sufficiently to ensure that non-senescent cells remain unstained.
